Sustained Release Compositions For Delivery Of Pharmaceutical Proteins

View page
US Patent:
20050203002, Sep 15, 2005
Filed:
Jan 7, 2005
Appl. No.:
11/031031
Inventors:
Stelios Tzannis - Newark CA, US
Nancy Dasovich - San Francisco CA, US
Sandeep Kumar - Thousand Oaks CA, US
Negar Sadrzadeh - San Carlos CA, US
International Classification:
A61K038/28
A61L009/04
A61K009/14
US Classification:
514004000, 424046000
Abstract:
The present invention provides sustained release compositions that can be manufactured easily while providing long duration of the pharmaceutically useful protein being delivered. The compositions comprise insoluble complexes of the pharmaceutically useful protein and a precipitating agent at an appropriate ratio to achieve a desired sustained release profile. The precipitating agent can be selected from a group of various agents including divalent metal cations, Hofmeister series salts and pH adjusters. Particularly provided are compositions comprising insulin that are suitable for pulmonary delivery.

Sustained Release Microparticles For Pulmonary Delivery

View page
US Patent:
20070031342, Feb 8, 2007
Filed:
May 9, 2006
Appl. No.:
11/430665
Inventors:
Stelios Tzannis - Newark CA, US
Negar Sadrzadeh - San Carlos CA, US
Helen Schiavone - Freeland WA, US
Renee Labiris - Paris, CA
International Classification:
A61L 9/04
A61K 38/23
A61K 31/573
US Classification:
424045000, 514012000, 514171000, 977906000
Abstract:
A composition of microparticles for delivery to the pulmonary system provides sustained release of a pharmaceutical agent. The microparticles comprise a lipid structural matrix comprising a multilamellar structure of lipid bilayers having lipid chains ordered in an Lphase. The lipid matrix at least partially encapsulates the pharmaceutical agent at a bilayer interface formed between head groups of adjacent lipid layers. The microparticles are prepared by heating a precursor formulation comprising a solvent, matrix-forming excipient and pharmaceutical agent to a temperature above the liquid-crystalline transition temperature Tof the matrix-forming excipient and below the melting or denaturation point of the pharmaceutical agent. The solvent is then removed to form microparticles with partially encapsulated pharmaceutical agent.
