Scientific Accomplishments Developed Rate Nephelometry assay for the quantitative analysis of Meninge A, C, W-135, & Y polysaccharides in the tetravalent polysaccharide mixture and for the bulk and final Meninge Infant protein-polysaccharide tetravalent vaccine product. Created developmental, optimization and validation documentation. Implemented and validated instrumentation as per IQ/OQ/PQ. Trained technicians on SWI and instrumentation post method transfer protocol. Developed liquid-liquid extraction process for the removal of residual phenol from Meninge IO In-process Animal-Free samples enabling BCA, O-Acetyl, Phosphorus and Sialic Acid assays to be used for in-process testing. Created development report and SWI. Trained technicians on SWI and safety considerations. Developed and implemented stability study for Meninge Infant polysaccharides A, C, W-135, & Y and protein-polysaccharide conjugates. Study included the testing of standards and controls against time and temperature crossed with lot variability. Created stability protocol and trained technicians. Co-developed qualitative protein assays for Diphtheria Toxoid analyses on both Beckman and Agilent CE systems using gel capillary electrophoresis. Attempted quantitation of Meninge Infant polysaccharides A, C, W-135, & Y using various tagging techniques. Co-developed robust assays for the following: Ethanol / acetone quantitation GLC , ammonium ion residual concentration Elite ion selective probe , protein concentration Menactra F.F.F. , Menomune kD related percent recovery testing HPSEC w / Millennium Package. Experienced with cGMP and ICH guidelines. Performed sample analysis of various products and intermediates with Finnegan ion trap mass spectrophotometer. Worked with FDA and Sanofi-Pasteur QC to remediate FDA PAl issue concerning BSA usage as standards and controls for release tests for final product(s). Developed testing scheme for BCA and Lowry assays using both BSA and Diphtheria Toxoid as standards and controls. Co-coordinated sample acquisition, performed sample dialysis, and assay sample preparation. Co-coordinated QC Total Nitrogen testing, followed by Lowry and BCA analyses. Compiled data with statistical analysis and interpretation. Presented to Sanofi-Pasteur AS&AD and QC lead scientists.
