Method For Controlling Streptococcus Pneumoniae Polysaccharide Molecular Weight Using Carbon Dioxide

View page
US Patent:
20100160622, Jun 24, 2010
Filed:
Dec 17, 2009
Appl. No.:
12/640666
Inventors:
Jean Heather Crinean - Sanford NC, US
Assignee:
Wyeth LLC - Madison NJ
International Classification:
C12P 19/04
C07H 11/04
US Classification:
536117, 435101
Abstract:
The present invention provides improved methods for producing a solution containing high molecular weight isolated capsular polysaccharides having phosphodiester linkages between saccharide repeat units. In certain methods, COis supplied to a fermentation culture of bacterial cells that produce capsular polysaccharide serotypes containing phosphodiester linkages between saccharide repeat units. Exemplary serotypes containing a phosphodiester linkage between saccharide repeat units include serotypes 6A, 6B, 19A, and 19F. Supplying COto the fermentation culture includes adding bicarbonate ions to the fermentation culture, adding carbonate ions to the fermentation culture, adding mixtures of bicarbonate and carbonate ions to the fermentation culture, and overlaying the fermentation culture with CO.

US Patent:
20100158953, Jun 24, 2010
Filed:
Dec 17, 2009
Appl. No.:
12/640769
Inventors:
Jean Heather Crinean - Sanford NC, US
Assignee:
Wyeth LLC - Madison NJ
International Classification:
A61K 39/09
A61P 31/04
A61P 37/04
C12P 19/04
US Classification:
4242441, 435101
Abstract:
The present invention provides improved methods for producing a solution containing high molecular weight isolated 19A capsular polysaccharides. In certain methods, a fermentation culture of bacterial cells that produce serotype 19A capsular polysaccharides is fermented for less than 6 hours before the bacterial cells are lysed the capsular polysaccharides are harvested. In other methods, COis supplied to the fermentation culture. Supplying COto the fermentation culture includes adding bicarbonate ions to the fermentation culture, adding carbonate ions to the fermentation culture, adding mixtures of bicarbonate and carbonate ions to the fermentation culture, and overlaying the fermentation culture with CO.
