Abstract:
The invention relates to phenol oxidizing enzymes encoded by nucleic acids capable of hybridizing to the nucleic acid having the sequence shown in SEQ ID NO: 1 and in particular phenol oxidizing enzymes obtainable from fungus. Abstract:
Disclosed herein are novel phenol oxidizing enzymes naturally-produced by strains of the species Stachybotrys which possess a pH optima in the alkaline range and which are useful in modifying the color associated with dyes and colored compounds, as well as in anti-dye transfer applications. Also disclosed herein are biologically-pure cultures of strains of the genus Stachybotrys, designated herein MUCL 38996 and MUCL 38898, which are capable of naturally-producing the novel phenol oxidizing enzymes. Disclosed herein is the amino acid and nucleic acid sequence for Stachybotrys phenol oxidizing enzymes as well as expression vectors and host cells comprising the nucleic acid. Disclosed herein are methods for producing the phenol oxidizing enzyme as well as methods for constructing expression hosts.

Abstract:
A method of increasing recovery of active enzyme produced in a plant is provided where the enzyme requires a transitional metal cofactor for activation. The metal cofactor is supplied to the enzyme during plant development, during extraction, or after extraction. Recovery of active enzyme is also provided by incubating the extracted enzyme at a non-enzyme degrading temperature. Addition of a negative ion salt further improves active enzyme recovery. Optimum salt concentrations for recovery of laccase from plants using copper solutions is provided.

Abstract:
The present invention provides variants of precursor phenol oxidizing enzymes. The precursor enzymes can be, for example, naturally-occurring or recombinant phenol oxidizing enzymes. The variant enzymes, in general, are obtained by modification of a precursor DNA sequence encoding the naturally-occurring or recombinant phenol oxidizing enzyme to generate the substitution of one or more amino acids in the encoded amino acid sequence. Variant enzymes are disclosed having properties which differ from those of their respective precursor enzymes, e. g. , altered pH optima and/or phenol oxidizing activity and/or altered stability. The variant enzymes of the present invention are especially useful in detergent formulations, for example, to modify (bleach) the color associated with colored compounds.
